چکیده

Benziman, Moshe (The Hebrew University of Jerusalem, Jerusalem, Israel), and Y. Galanter. Flavine adenine dinucleotide-linked malic dehydrogenase from Acetobacter xylinum. J. Bacteriol. 88:1010-1018. 1964.-The properties of the pyridine nucleotide-nonlinked malic dehydrogenase of Acetobacter xylinum were investigated in the supernatant fluid obtained by high-speed centrifugation of sonic extracts. Ferricyanide, phenazine methosulfate, and to a lesser extent dichlorophenolindophenol were active as oxidants for malate oxidation. After acid ammonium sulfate precipitation, the enzyme lost its malate-oxidizing activity. The enzyme was reactivated by low concentrations of flavine adenine dinucleotide (FAD) but not by flavine mononucleotide (FMN) or riboflavine. Atabrine inhibited the enzyme, and the inhibition was relieved by FAD but not by FMN or riboflavine. Malate-oxidizing activity was inhibited by hematin. The inhibition was prevented by imidazole or globin. o-Phenanthroline, 8-hydroxy quinoline, alpha,alpha'-dipyridyl, and p-chloromercuribenzoate inhibited malate oxidation. Amytal markedly inhibited oxidation of malate in the presence of oxygen, phenazine methosulfate, or dichlorophenolindophenol, but not in the presence of ferricyanide. The results suggest that the malic dehydrogenase of A. xylinum is a FAD enzyme, which contains an ironbinding site essential for its activity. Nonheme iron and sulfhydro groups are possibly involved in enzyme activity. The malic dehydrogenase is functionally linked to the cytochrome chain.

a+Ketoglutarate Dehydrogenase Complex of Acetobacter xylinum

The cu-ketoglutarate dehydrogenase complex of Acetobacterxylinum was purified to homogeneity. It consists of three main polypeptide chains with a total molecular weight of about 2.4 x 106. It catalyzes the overall Mg*+ and thiamin pyrophosphate-dependent, NAD+and CoA-linked oxidative decarboxylation of cY-ketoglutarate, as well as the partial reactions characteristic of the three enzyme compone...

Regulatory Properties of the a-Ketoglutarate Dehydrogenase Complex of Acetobacter xylinum

The initial velocity studies on the a-ketoglutarate dehydrogenase complex in toluene/Triton X-lOO-permeabilized Acetobwter xylinum cells revealed that the regulatory properties of the complex, in situ, are the same as for the purified complex. Thus, sigmoidal kinetics with respect to a-ketoglutarate were obtained (So.5 (a-ketogkltarate) = 5 lfl~ ; nH = 1.8), which convert into a hyperbolic rela...

The Effect of Adenylic Acid on Yeast Nicotinamide Adenine Dinucleotide Isocitrate Dehydrogenase, a Possible Metabolic Control Mechanism.

The nicotinamide adenine dmucleotide-specific isocitrate dehydrogenase of yeast was reported by Kornberg and Pricer (1) to have an absolute requirement for adenylic acid and not to catalyze the reverse reaction under conditions suitable for such catalysis by the nicotinamide adenine dinucleotide phosphatespecific enzyme. Similar results have been obtained for the nicotinamide adenine dinucleoti...

Influence of carboxylic acids on the stereospecific nicotinamide adenine dinucleotide-dependent and nicotinamide adenine dinucleotide-independent lactate dehydrogenases of Leuconostoc mesenteroides.

Leuconostoc mesenteroides increased its lactic acid production from glucose threefold when malic acid was added to the culture. This increase resulted also in a reduction of the ratio of d-lactic acid to l-lactic acid (31.5 to 1.23). Addition of malic acid increased 6.5-fold the specific activity of nicotinamide adenine dinucleotide (NAD)-linked l-lactate dehydrogenase and increased 3.2-fold th...
